Review on Buccal Mucoadhesive Drug Delivery System in the Human Body

Abstract
Considering its ease of administration, the oral cavity is a preferred place for pharmaceutical distribution. This approach enables the delivery of medications both mucosally (local effect) and transmucosally (systemic effect). The goal in the first case is to achieve a site-specific release of the drug on the mucosa, whereas in the second case, the drug is absorbed through the mucosal barrier and into the systemic circulation. The primary obstacles that drugs confront when administered bucally are the narrow absorption area and mucosal barrier properties. Other obstacles to consider include the oral cavity’s superior physiological clearance processes, which move the formulation away from the absorption site. Using new materials with mucoadhesive, enzyme inhibitory, and permeation-enhancing properties, to overcome these limitations, researchers are exploring innovative drug delivery technologies that increase patient compliance and promote closer contact between the drug and the absorption mucosa. This overview covers the benefits and downsides of buccal medication delivery, including the anatomy of the oral mucosa, drug permeation mechanisms, current formulation design, and improvements in buccal delivery methods.
